=== Pharmacokinetics === Ceftolozane–tazobactam is available as a 2:1 fixed combination (such that a 1.5 g dose of ceftolozane–tazobactam is composed of 1 g of ceftolozane and 500 mg of tazobactam). Ceftolozane-tazobactam is administered intravenously. For both ceftolozane and tazobactam, the peak plasma concentration occurs immediately after a 60 minute infusion, with a time to maximum concentration of approximately one hour. The binding of ceftolozane to human plasma proteins is approximately 16% to 21%, while the binding of tazobactam is approximately 30%. The mean steady-state volume of distribution in healthy adult males after a single 1.5 g IV dose is 13.5 L for ceftolozane and 18.2 L for tazobactam, which is similar to extracellular fluid volume. Tissue distribution of ceftalozone-tazobactam is rapid and shows good penetration into the lung, rendering it an ideal treatment for bacterial pneumonia. The metabolism and excretion of ceftolozane are similar to those of most β-lactam antimicrobial agents. Ceftolozane is not metabolized to any significant extent and thus predominantly eliminated unchanged in the urine. Tazobactam is partially metabolized to an inactive metabolite, and both drug and metabolite are excreted in the urine (80% as unchanged drug). == Enrichment steps == The peptide-mRNA:cDNA fusions can be selected over immobilized selection targets for several rounds (Figure 3). There might be a relatively high background for the first few rounds of selection, and this can be minimized by increasing selection stringency, such as adjusting salt concentration, amount of detergent, and/or temperature during the target/fusion binding period. Following binding selection, those library members that stay bound to the immobilized target are PCR amplified. The PCR amplification step will enrich the population from the mRNA-display library that has higher affinity for the immobilized target. Error-prone PCR can also be done in between each round of selection to further increase the diversity of the mRNA-display library and reduce background in selection. === Predators === Newborn rattlesnakes are heavily preyed upon by a variety of species, including cats, ravens, crows, roadrunners, raccoons, opossums, skunks, coyotes, weasels, whipsnakes, kingsnakes, and racers. Young of the smaller crotaline species are frequently killed and eaten by small predatory birds, such as jays, kingfishers, and shrikes. Some species of ants in the genus Formica are known to prey upon neonates, and Solenopsis invicta (fire ants) likely do, as well. On occasion, hungry adult rattlesnakes cannibalize neonates. The small proportion (often as few as 20%) of rattlesnakes that make it to their second year are heavily preyed upon by a variety of larger predators, including coyotes, eagles, hawks, owls, falcons, feral pigs, badgers, indigo snakes, and kingsnakes. The common kingsnake (Lampropeltis getula), a constrictor, is immune to the venom of rattlesnakes and other vipers, and rattlesnakes form part of its natural diet. Rattlesnakes sense kingsnakes' presence by their odor. When they realize a kingsnake is nearby, they begin enacting a set of defensive postures known as "body bridging". Unlike its normal erect and coiled defensive-striking posture, the rattlesnake keeps its head low to the ground in an attempt to prevent the kingsnake from gaining a hold on it (the head being the first part of the rattlesnake to be ingested). The rattlesnake jerks its body about, while bridging its back upwards, forming an elevated coil that faces the kingsnake. The elevated coil is used to strike the attacker, and is also used to shield the head from the kingsnake.
=== Relations with Tibet === In the fall of 1933, the 13th Dalai Lama sent an ultimatum to Liu Wenhui stating that Tibet would launch another attack if the 24th Army did not retreat further east. The ultimatum was not followed up because of the Lama's death on 17 December. In the wake of the ensuing political instability within the region, Liu cabled Nanjing for an expeditionary force to restore Chinese control over the entirety of Tibet. However, Dai Jitao and other close advisors to Chiang Kai-shek chose a more conciliatory policy and urged restraint on 23 December, which Liu Wenhui again ignored. Here, historian James Leibold speculates that Liu would probably have seized Lhasa had it not been for his untimely defeat at the hands of his nephew. In 1934, the new Tibetan government of Jamphel Yeshe Gyaltsen was ready to pursue peace with the central government, as the war with Liu had drained the monasteries' resources and driven up the price of Chinese tea. Chiang dispatched General Huang Musong on a diplomatic mission to Lhasa, with the aim of getting Tibet to accept Chinese sovereignty in exchange for broad autonomy. This mission, although accepted by the Kashag, was foiled when Liu Wenhui met with Huang in Chengdu and urged a hard line on the border issue. This was anathema to the Tibetans who wanted a demilitarized buffer zone in Xikang to protect from Liu Wenhui's army.
