This is a working overview of electrospray ionisation, written for readers who want more than a one-paragraph summary but less than a textbook.
This page was last updated on 2026-08-01 and is reviewed periodically as new material appears.
Identity and purity are checked by reversed-phase high-performance liquid chromatography, usually with ultraviolet detection near 214 nanometres, where the peptide bond absorbs. Mass spectrometry confirms the expected mass and reveals whether truncation products or adducts are present. Acid hydrolysis followed by amino acid analysis gives the residue ratio, which should approximate one alanine, one glutamate, one aspartate and one glycine. Counter-ions such as acetate or trifluoroacetate remain in the dried product and lower net peptide content, so a stated purity figure on a label does not by itself describe how much peptide a vial holds.
Regulatory treatment differs by country. No formulation of epitalon holds a marketing authorisation as a medicine in the United States or the European Union, where material sold for laboratory use is handled as a research chemical and is not intended for human consumption. In Russia, several short peptide preparations from the same institute's peptide series are registered medicinal products, and epitalon appears in that national context. Elsewhere it is frequently offered as a cosmetic ingredient, a category with lighter requirements. Advertising claims about longevity or disease prevention are restricted in most jurisdictions, which limits how sellers describe the compound.
Human data are limited to small studies, often without the randomization, blinding, or control groups expected in contemporary clinical research. Reported outcomes have included changes in melatonin levels, immune markers, and subjective measures, but sample sizes were generally too small to support firm conclusions. Some reviews treat the peptide as promising while noting methodological weaknesses; others question whether the observed effects are specific. The compound is frequently discussed in longevity-focused communities, where enthusiasm often outpaces the published evidence. Separating established findings from speculation is therefore important when reading summaries of this research.
Epitalon emerged from research conducted in Saint Petersburg by Vladimir Khavinson and colleagues, who studied short peptides as potential regulators of aging. The work built on epithalamin, a pineal gland extract reported to influence neuroendocrine function. Epitalon was designed as a synthetic counterpart with a defined sequence, allowing reproducible experiments that extracts could not support. Early publications described effects on melatonin rhythms and lifespan in animal models. These findings circulated mainly in Russian-language journals during the 1990s, which limited their visibility among English-speaking researchers.
The most widely cited claim is that epitalon activates telomerase and thereby extends telomere length. Supporting evidence comes largely from cultured human cells, where treatment was associated with increased telomerase activity and delayed replicative senescence. Telomerase activation is a biologically consequential effect, since the enzyme is largely silenced in most somatic cells. However, the route by which a short peptide would reach and act on the enzyme's regulatory machinery has not been established. Independent replication in human trials is scarce, so the link between cell-culture observations and whole-body aging remains an open question.

No pharmacopoeial monograph exists for this peptide, so quality rests on the supplier's internal specifications and on whatever independent testing a purchaser arranges. Certificates of analysis differ widely in which tests they report and in the limits applied. The counter-ion introduced during purification, commonly acetate or trifluoroacetate, changes the net peptide content of a given mass of powder, so two samples of equal weight may not contain equal amounts of the active sequence. Third-party laboratories can verify purity, identity, residual solvents, and counter-ion content for a fee, which makes documentation more informative than labelling.
Verification of a sample usually begins with reversed-phase high-performance liquid chromatography, which resolves the peptide from truncated sequences and other impurities and expresses purity as a percentage of total peak area. Mass spectrometry by electrospray ionisation or matrix-assisted laser desorption supplies an independent check, because the measured mass can be compared against the theoretical value for AEDG. Amino acid analysis or tandem mass spectrometry sequencing can confirm residue order. Each of these methods answers a different question: a purity figure does not establish identity, and an identity match does not establish how much of the material is intact peptide.
Purity assessment of peptide reagents normally relies on reversed-phase high-performance liquid chromatography. A gradient of acetonitrile in water with trifluoroacetic acid, paired with a C18 stationary phase, separates the target tetrapeptide from truncated sequences and deletion analogues. Detection at 214 nanometres exploits absorbance of the peptide backbone, since the molecule contains no aromatic residue. Results are expressed as a percentage of total peak area. Values above ninety-five percent are typical for research-grade material, although reporting conventions vary between suppliers.
Identity confirmation uses mass spectrometry, usually electrospray ionisation in positive mode or matrix-assisted laser desorption. The protonated free peptide produces a signal near three hundred and ninety-one daltons, while salt adducts shift that value slightly. Tandem mass spectrometry or amino acid analysis supplies sequence-level confirmation, which a single mass measurement cannot. Peptides containing aspartate can cyclise into succinimide intermediates that hydrolyse to isoaspartate isomers of identical mass. Because those isomers are difficult to separate chromatographically, identity and purity results carry more weight when interpreted together.
Storage practice centres on limiting moisture, oxygen, and repeated temperature cycling. Freeze-dried powder is generally held desiccated at minus twenty degrees Celsius or colder and protected from light. Reconstituted solutions are markedly less stable, and the aspartate-glycine junction is prone to succinimide formation at neutral to mildly alkaline pH. Portioning material into single-use aliquots reduces degradation compared with repeated freeze-thaw cycles. Stability data specific to this tetrapeptide remain scarce, so most handling guidance is extrapolated from general peptide chemistry rather than measured directly.
This loop is believed to play an important role in stabilizing the cyclotide structure through hydrogen bonding with residues from loops 3 and 5. Loops 2-6 also have highly conserved features, including the ubiquitous presence of just a single amino acid in loop 4 that is likely involved in sidechain-sidechain hydrogen bonding. Other conserved residues include a hydroxyl-containing residue in loop 3, a glycine residue in the final position of loop 3, a basic and a proline residue in the penultimate position in loop 5 of bracelet and Möbius cyclotides respectively, and an asparagine (or occasionally aspartic acid) residue at the putative cyclisation point in loop 6. It is of interest to note that not only are certain residues highly conserved, but the backbone and side chain angles are as well. With recent screening programs suggesting that the number of cyclotide sequences may soon reach the thousands, a database, CyBase, has been developed that offers the opportunity for comparisons of sequences and activity data for cyclotides. Several other families of circular proteins are known in bacteria, plants and animals and are also included in CyBase.
=== Blood pressure === Serum vitamin C was reported to be 15.13 μmol/L lower in people with hypertension compared to normotensives. The vitamin was inversely associated with both systolic blood pressure (SBP) and diastolic blood pressure (DBP). Oral supplementation of the vitamin resulted in a modest but statistically significant decrease in SBP in people with hypertension. The proposed explanation is that vitamin C increases intracellular concentrations of tetrahydrobiopterin, an endothelial nitric oxide synthase cofactor that promotes the production of nitric oxide, which is a potent vasodilator. Vitamin C supplementation might also reverse the nitric oxide synthase inhibitor NG-monomethyl-L-arginine 1, and there is also evidence cited that vitamin C directly enhances the biological activity of nitric oxide.
The main effect of glaucoma is damage to the optic nerve. Eventually, this damage leads to vision loss, which can deteriorate with time. The underlying cause of open-angle glaucoma remains unclear. Several theories exist on its exact etiology. Intraocular pressure is a function of the production of liquid aqueous humor by the ciliary processes of the eye, and its drainage through the trabecular meshwork. Aqueous humor flows from the ciliary processes into the posterior chamber, bounded posteriorly by the lens and the zonules of Zinn, and anteriorly by the iris. It then flows through the pupil of the iris into the anterior chamber, bounded posteriorly by the iris and anteriorly by the cornea. From here, the trabecular meshwork drains aqueous humor via the scleral venous sinus (Schlemm's canal) into scleral plexuses and general blood circulation. In open/wide-angle glaucoma, flow is reduced through the trabecular meshwork due to the degeneration and obstruction of the trabecular meshwork, whose original function is to absorb the aqueous humor. Loss of aqueous humor absorption leads to increased resistance and thus a chronic, painless buildup of pressure in the eye. In primary angle-closure glaucoma, the iridocorneal angle is narrowed or completely closed, obstructing the flow of aqueous humor to the trabecular meshwork for drainage. This is usually due to the forward displacement of the iris against the cornea, resulting in angle closure. This accumulation of aqueous humor causes an acute increase in pressure and damage to the optic nerve.
=== Diabetes-related modifiers of cellular function === Necrobiosis Lipoidica commonly co-occurs with Type 1 diabetes. The metabolic dysregulation found in diabetes has a profound impact on cellular function. Diabetes is known to alter the expression of miRNAs and promote oxidative stress. Prolonged high blood sugar can increase oxidative stress, disrupt nitric oxide production, essential for vasodilation and vessel formation. Hyperglycemia also leads to the accumulation of advanced glycation end-products (AGEs), which activate the RAGE–NF-κB axis, leading to inflammation and endothelial apoptosis. Structural changes occur, including basement membrane thickening and extracellular matrix stiffening.
=== Hygiene hypothesis === According to the hygiene hypothesis, high levels of cleanliness expose children to fewer antigens than in the past, causing their immune systems to become overactive and more likely to misidentify own tissues as foreign, resulting in autoimmune or allergic conditions such as asthma.

Components of the ECM are produced intracellularly by resident cells and secreted into the ECM via exocytosis. Once secreted, they then aggregate with the existing matrix. The ECM is composed of an interlocking mesh of fibrous proteins and glycosaminoglycans (GAGs).
Because of thorium's radioactivity, minerals containing it are often metamict (amorphous), their crystal structure having been damaged by the alpha radiation produced by thorium. An extreme example is ekanite, (Ca,Fe,Pb)2(Th,U)Si8O20, which almost never occurs in nonmetamict form due to the thorium it contains. Monazite (chiefly phosphates of various rare-earth elements) is the most important commercial source of thorium because it occurs in large deposits worldwide, principally in India, South Africa, Brazil, Australia, and Malaysia and is mined for its rare earth content. It contains around 2.5% thorium on average, although some deposits may contain up to 20%. Monazite is a chemically unreactive mineral that is found as yellow or brown sand; its low reactivity makes it difficult to extract thorium from it. Allanite (chiefly silicates-hydroxides of various metals) can have 0.1–2% thorium and zircon (chiefly zirconium silicate, ZrSiO4) up to 0.4% thorium. Thorium dioxide occurs as the rare mineral thorianite. Due to its being isotypic with uranium dioxide, these two common actinide dioxides can form solid-state solutions and the name of the mineral changes according to the ThO2 content. Thorite (chiefly thorium silicate, ThSiO4), also has a high thorium content and is the mineral in which thorium was first discovered. In thorium silicate minerals, the Th4+ and SiO4−4 ions are often replaced with M3+ (where M = Sc, Y, or Ln) and phosphate (PO3−4) ions respectively.
=== Mandatory minimums === The Anti-Drug Abuse Act of 1986 established a 100-1 sentencing disparity for the possession of crack or powder cocaine. Possession of 500 g of powder cocaine triggered a five-year mandatory minimum sentence, but it took possession of 5 g of crack cocaine to trigger the same mandatory minimum penalty. In addition, the Anti-Drug Abuse Act of 1988 established a one-year mandatory minimum penalty for simple possession of crack cocaine, which made crack cocaine the only controlled substance for which a first possession offense triggered a mandatory minimum penalty. A 1992 study found that mandatory minimum sentencing caused blacks and Hispanics to receive more-severe sentences than their white counterparts from 1984 to 1990. In 1995, the United States Sentencing Commission delivered a report to Congress concluding that because 80% of crack offenders were black, the 100–1 disparity disproportionately affected minorities. It recommended for the crack-powder sentencing ratio to be amended, and other sentencing guidelines to be re-evaluated. Its recommendations were rejected by Congress. By contrast, certain authors have pointed out that the Congressional Black Caucus backed the Anti-Drug Abuse Act of 1986, which implied that that law could not be racist. In 2010, Congress passed the Fair Sentencing Act, which reduced the sentencing disparity between crack and powder cocaine from 100–1 to 18–1. The mandatory minimum penalty was amended to take effect for possession of crack cocaine in excess of 28 g.
FC SKChF Sevastopol (reconstituted based on FC Sevastopol) FC TSK Simferopol (reconstituted based on SC Tavriya Simferopol) FC Zhemchuzhina Yalta (revived based on same club that was disqualified from Ukrainian competitions) The three clubs are from Crimea, a territory recognized by Ukraine and a majority of countries as part of Ukraine, but have been under effective Russian control as the Republic of Crimea since the annexation of Crimea by the Russian Federation. FC Sevastopol and SC Tavriya Simferopol last played in the 2013–14 Ukrainian Premier League, and were dissolved after the completion of the season. The three clubs were reformed as football organizations of the Russian Federation and joined the Russian Professional Football League starting from the 2014–15 season, after approval from the Russian Football Union. The inclusion of Crimean clubs in Russian competitions have not been approved by either FIFA or UEFA, and the Football Federation of Ukraine have lodged a complaint. On 22 August 2014, UEFA decided "that any football matches played by Crimean clubs organised under the auspices of the Russian Football Union will not be recognised by UEFA until further notice", and on 4 December 2014, decided to prohibit Crimean clubs to play in competitions organised by the Russian Football Union as from 1 January 2015 and for the region to be considered as a "special zone" for football purposes until further notice. In 2023 Crimean clubs began playing in the Russian system again, despite being a UEFA "special zone".
Nearly two billion people in the developing world are deficient in zinc. Groups at risk include children in developing countries and the elderly with chronic illnesses. In children, it causes an increase in infection and diarrhea and contributes to the death of about 800,000 children worldwide per year. The World Health Organization advocates zinc supplementation for severe malnutrition and diarrhea. Zinc supplements help prevent disease and reduce mortality, especially among children with low birth weight or stunted growth. However, zinc supplements should not be administered alone, because many in the developing world have several deficiencies, and zinc interacts with other micronutrients. While zinc deficiency is usually due to insufficient dietary intake, it can be associated with malabsorption, acrodermatitis enteropathica, chronic liver disease, chronic renal disease, sickle cell disease, diabetes, malignancy, and other chronic illnesses. In the United States, a federal survey of food consumption determined that for women and men over the age of 19, average consumption was 9.7 and 14.2 mg/day, respectively. For women, 17% consumed less than the EAR, for men 11%. The percentages below EAR increased with age. The most recent published update of the survey (NHANES 2013–2014) reported lower averages – 9.3 and 13.2 mg/day – again with intake decreasing with age. Symptoms of mild zinc deficiency are diverse.

=== Other === Hydrogels also possess a degree of flexibility very similar to natural tissue due to their significant water content. As responsive "smart materials", hydrogels can encapsulate chemical systems which upon stimulation by external factors such as a change of pH may cause specific compounds such as glucose to be liberated to the environment, in most cases by a gel–sol transition to the liquid state. Chemomechanical polymers are mostly also hydrogels, which upon stimulation change their volume and can serve as actuators or sensors.
=== Fish === The skeleton, which forms the support structure inside the fish is either made of cartilage as in the Chondrichthyes, or bones as in the Osteichthyes. The main skeletal element is the vertebral column, composed of articulating vertebrae which are lightweight yet strong. The ribs attach to the spine and there are no limbs or limb girdles. They are supported only by the muscles. The main external features of the fish, the fins, are composed of either bony or soft spines called rays which, with the exception of the caudal fin (tail fin), have no direct connection with the spine. They are supported by the muscles which compose the main part of the trunk. Cartilaginous fish, such as sharks, rays, skates, and chimeras, have skeletons made entirely of cartilage. The lighter weight of cartilage allows these fish to expend less energy when swimming.

== Description == The cap is white, may have fine scales, and is 3 to 12 centimetres (1+1⁄4 to 4+3⁄4 inches) in diameter; it is first hemispherical in shape before flattening out with maturity. The gills are initially pink, then red-brown and finally a dark brown, as is the spore print. The stipe is 3 to 10 cm (1+1⁄4 to 4 in) tall, 1–2 cm wide, predominantly white and bears a single thin ring. The taste is mild. The white flesh bruises a dingy reddish brown, as opposed to yellow in the poisonous Agaricus xanthodermus and similar species. The thick-walled, dark brown, elliptical spores measure 5.5–8 μm by 4–5 μm. Cheilocystidia are absent.

The solid is kept cold, dry and dark, typically at minus twenty degrees Celsius, in a sealed container with desiccant. Dissolved material is kept refrigerated and used promptly. Freezing and thawing a solution repeatedly is avoided because it accelerates degradation.
Reversed-phase chromatography with ultraviolet detection gives a purity percentage, while mass spectrometry confirms identity. Amino acid analysis checks the residue composition. Buyers typically request a certificate of analysis showing these measurements for the specific lot.
It is not authorised as a medicine in the United States or the European Union. Material is sold there as a research chemical or cosmetic ingredient. Some related short peptides are registered drugs in Russia, which is a distinct national situation rather than a general approval.
Vladimir Khavinson and colleagues at research institutes in Saint Petersburg developed and studied the peptide. Their program examined short peptides as regulators of aging and neuroendocrine function. Epitalon was one of several compounds produced by that group.
